Can we predict protein function from structure?

AlphaFold and its successors have largely solved structure prediction from amino-acid sequence. Predicting the biochemical function of a structure — and how it interacts with other proteins in a cellular context — is a harder open problem.

Progress requires combining structural biology with high-throughput perturbation experiments, machine learning on interaction data, and better models of post-translational regulation.
